No left or right to the "clamshell" mounts on the block, but if the rubber is wore out in the rubber-encapsulated frame side mount it can give you problems with alignment.

The haynes manual shows two different mounts, but I just replaced the 350 in my 83 P/U with a 454 and it went right in. The last time I did this swap, I bought the "BB" mounts thinking they were different but It appears the stock ones work.

Have you slid the transmission back as far as possible? Loosen but don't remove the transmission to crossmember fastener, and slide it back towards the bed. Should provide some additional clearance.
