Can I do this?

I need to bush down a out port on a duel master cylinder. Can I use a standard NPT bushing. The 90 degree 3/16" brake line fitting will then screw in. Will the thing leak beacuse the fittings, especially the bushing are not making sealing contact to the master cylinder flare?

yes it will leak plus the threads wont match.

NAPA sells the correct adapter fittings for different size inverted flare fittings.
